用氧化型低密度脂蛋白处理培养的猪主动脉内皮细胞,以Ficoll-Hapaque分离液密度梯度离心分离人外周血单核细胞,观察L-精氨酸对内皮细胞-单核细胞粘附率的影响;采用反转录-多聚酶链反应技术检测氧化型低密度脂蛋白和L-精氨酸对内皮细胞血管细胞粘附分子-1表达的影响。结果显示:L-精氨酸具有剂量和时间依赖性地抑制氧化型低密度脂蛋白的促内皮细胞-单核细胞粘附作用。氧化型低密度脂蛋白处理内皮细胞能明显增加内皮血管细胞粘附分子。-1的表达,而L-精氨酸阻抑氧化型低密度脂蛋白的此种作用。结果提示L-精氨酸可能通过抑制血管细胞粘附分子-1表达阻抑氧化型低密度脂蛋白的促内皮细胞—单核细胞粘附作用。
Cultured porcine aortic endothelial cells were treated with oxidized low density lipoprotein and human peripheral blood mononuclear cells were isolated by Ficoll-Hapaque separation gradient centrifugation to observe the effect of L-arginine on endothelial cell-monocyte adhesion rate The effects of oxidized low density lipoprotein and L-arginine on the expression of vascular cell adhesion molecule-1 in endothelial cells were detected by reverse transcription polymerase chain reaction (RT-PCR). The results showed that L-arginine could inhibit EPCs-monocyte adhesion of oxidized low density lipoprotein in a dose-and time-dependent manner. Oxidized low-density lipoprotein-treated endothelial cells significantly increased endothelial cell adhesion molecules. -1 expression, whereas L-arginine suppresses this effect of oxidized low density lipoprotein. The results suggest that L-arginine may inhibit the endothelial cell-monocyte adhesion of oxidized low-density lipoprotein by inhibiting the expression of vascular cell adhesion molecule-1.
