目的通过体内和体外模型研究知母皂苷对芒果苷肠吸收的影响。方法采用液-质联用法测定大鼠灌胃芒果苷、知母皂苷-芒果苷后的血药浓度,并计算主要药动学参数;应用大鼠肠外翻模型,UPLC法测定不同时间点时肠囊内芒果苷的浓度,计算肠吸收参数。结果合用知母皂苷后,芒果苷的血药浓度比单体组明显升高,药时曲线下面积(AUC)和Cmax显著增加,Tmax延长至3 h;在回肠段知母皂苷-芒果苷(1∶1)组中芒果苷的吸收参数Q90、Vt和Paap均高于单体组,但差异无统计学意义(P>0.05)。结论知母皂苷对芒果苷的体内吸收有一定的促进作用。
Objective To study the effect of timosaponins on intestinal absorption of mangiferin by in vivo and in vitro models. Methods The plasma concentrations of mangiferin, timosaponin-mangiferin in rats were determined by liquid chromatography-mass spectrometry and the main pharmacokinetic parameters were calculated. At the different time points Intestinal capsule mangiferin concentration, calculate intestinal absorption parameters. The results of the combination of timosaponins, the plasma concentration of mangiferin was significantly higher than the monomer group, the drug area under the curve (AUC) and Cmax increased significantly, Tmax extended to 3 h; in the ileum timosaponin - mangiferin ( 1: 1) group, the absorption parameters of mangiferin, Q90, Vt and Paap were higher than those of the monomer group, but the difference was not statistically significant (P> 0.05). Conclusion Inhaled saponin Mangiferin in vivo absorption has a certain role in promoting.
