目的:探讨连接蛋白(Cx)基因对人脑胶质瘤细胞生长的抑制作用。方法:以脂质体介导将Cx43cDNA导入Cx43表达缺失的TJ905人胶质母细胞瘤细胞;Northern杂交、原位杂交及免疫组化检测Cx43mRNA及蛋白表达;MTT法和AgNOR染色检测细胞增殖;TUNEL法测细胞凋亡。结果:转染后TJ905细胞有Cx43mRNA及蛋白表达,高表达的克隆增殖明显下降。细胞凋亡不增加。结论:Cx43基因可能成为恶性胶质瘤基因治疗的优选靶之一。
Objective: To investigate the inhibitory effect of connexin (Cx) gene on the growth of human glioma cells. Methods: The Cx43 cDNA was introduced into TJ905 human glioblastoma cells without Cx43 expression by liposome. Northern blotting, in situ hybridization and immunohistochemistry were used to detect the expression of Cx43 mRNA and protein. Cell proliferation was detected by MTT assay and AgNOR staining. TUNEL Law measured apoptosis. Results: After transfected TJ905 cells with Cx43 mRNA and protein expression, the high expression of the clone proliferation decreased significantly. Apoptosis does not increase. Conclusion: Cx43 gene may be one of the preferred targets for gene therapy of malignant glioma.
