目的:探讨活血、利水中药对脑出血大鼠蛋白酶激活受体-1(PAR-1)表达的影响。方法:168只雄性SD大鼠随机分为假手术组、模型组、活血组和利水组。采用自体不凝血注入尾状核法制作大鼠脑出血模型,分别于术后1、3、5d 3个时间点取材,组织干湿重法计算脑含水量,免疫组织化学法和Real-time PCR法检测脑组织PAR-1蛋白和基因的表达。结果:与假手术组相比,模型组各时间点大鼠脑组织含水量增多(P<0.01),PAR-1蛋白和基因的表达均明显升高(P<0.01);活血组和利水组大鼠脑组织含水量及PAR-1的表达较模型组降低(P<0.05),且活血方优于利水方(P<0.05)。结论:活血、利水中药均能减轻脑出血后脑水肿,其机制可能与降低PAR-1表达有关。
Objective: To investigate the effects of Huoxue and Lishui herbs on protease-activated receptor-1 (PAR-1) expression in rats with intracerebral hemorrhage. Methods: 168 male SD rats were randomly divided into sham-operation group, model group, Huoxue group and Li-shui group. The rat model of intracerebral hemorrhage was made by autologous non-coagulated injection of caudate nucleus. The rats were sacrificed at 1, 3 and 5 days after operation. The brain water content was calculated by the wet-dry method. Immunohistochemistry and Real-time PCR Method to detect the expression of PAR-1 protein and gene in brain tissue. Results: Compared with sham operation group, the water content of brain tissue increased (P <0.01) and the expression of PAR-1 protein and gene in model group increased significantly at each time point (P <0.01) The water content of brain tissue and the expression of PAR-1 in rat brain were lower than those in model group (P <0.05). Conclusion: Both Huoxue and Lishui herbs can relieve cerebral edema after intracerebral hemorrhage. The mechanism may be related to the decrease of PAR-1 expression.
