目的通过研究消银解毒饮及其拆方对角质形成细胞COLO-16分泌血管内皮长生因子(VEGF)的影响,探讨消银解毒饮治疗银屑病的作用机制。方法将36只SD大鼠随机分为消银解毒饮组、祛风除湿方组、解毒方组、凉血方组、甲氨喋呤组及对照组,给药5d后取各组大鼠含药血清,加入培养好的角质形成细胞COLO-16中,用双抗体夹心酶联免疫法测定角质形成细胞分泌VEGF的含量。结果与对照组比较VEGF含量,在5%浓度时,消银解毒饮血清组和甲氨喋呤血清组均明显下降,以消银解毒饮血清组下降显著(P<0.01);在10%浓度时,消银解毒饮血清组、解毒方血清组及甲氨喋呤血清组均明显下降,以消银解毒饮血清组和解毒方血清组下降显著(P<0.01);在20%浓度时,消银解毒饮血清组、解毒方血清组、凉血方血清组以及甲氨喋呤血清组均明显下降,以消银解毒饮血清组、解毒方血清组及甲氨喋呤血清组下降显著(P<0.01)。结论消银解毒饮及其拆方除祛风除湿方外,其余几种含药血清对COLO-16细胞分泌VEGF均有抑制作用,解毒药物对VEGF的分泌发挥主要的抑制作用;抑制角质形成细胞分泌VEGF,可能是消银解毒饮抑制血管内皮细胞增生,治疗银屑病的又一重要作用机制。
Objective To study the effect of Xiaoyin Jiedu Decoction and its decomposed prescriptions on the secretion of vascular endothelial growth factor (VEGF) from keratinocytes (COLO-16) and to explore the mechanism of Xiaoyinjieduyin in the treatment of psoriasis. Methods Thirty-six SD rats were randomly divided into Xiaoyin Jiedu Yin group, Qufeng Qushi Decoction group, Jiedufang group, Liangxue Fang group, Methotrexate group and control group. Rats in each group were given 5 days after administration. Drug serum was added to cultured keratinocyte COLO-16, and the VEGF content in keratinocytes was measured by double-antibody sandwich enzyme-linked immunosorbent assay. Results Compared with the control group, the VEGF level was significantly decreased at the 5% concentration in the Xiaoyin Jiedu Yin group and the methotrexate serum group, and was significantly decreased in the Xiaoyin Jiedu Yin group (P<0.01); At the same time, Xiaoyinjieduyin serogroup, detoxification serogroup and methotrexate serogroup were all significantly decreased, with a significant decrease in Xiaoyinjieduyin serogroup and Jiedufang serogroup (P<0.01); at 20%, The serum levels of Xiaoyin Jieduyin Decoction, Jiedufang Decoction Group, Liangxuefang Decoction Group, and Methotrexate Serum Deposition Group all decreased significantly. The serum levels of Xiaoyinjiedu Decoction, Detoxification Detoxification Serum, and Methotrexate Serum Degraded significantly ( P<0.01). [Conclusion] Xiaoyin Jiedu Decoction and its decomposed prescriptions except decoction and dehumidifier, other kinds of drug-containing serum can inhibit the secretion of VEGF in COLO-16 cells, and detoxification drugs play a major role in inhibiting the secretion of VEGF; inhibiting keratinocytes Secretion of VEGF may be another important mechanism for the treatment of psoriasis by inhibiting the proliferation of vascular endothelial cells.
