本文利用SDS-PAGE、Westernblotting.DotELISA等分子流行病学方法分析了21株不同地区、不同来源的中国莱姆病螺旋体菌株的蛋白组成及单克隆抗体反应性。研究发现中国菌株主要蛋白呈现高度多态性和异质性,主要外膜蛋白OspA、OspB及PC蛋白有数种不同的表型,并呈现出紧密的相关性。所有的菌株均与单克隆抗体H9724(抗41KO鞭毛抗原)呈阳性反应。除几株菌株外,绝大多数中国菌株均不与H6831(抗OspB单克隆抗体)反应。中国菌株与单克隆抗体H5332(抗OspA单克隆抗体)呈现不同的反应性,有强、弱和无反应等。根据中国菌株的蛋白组成及单克隆抗体反应性可大致将其分成三组,其中第Ⅰ组菌株具有与北美代表株B31完全一致的蛋白组成及单克隆抗体反应性。第Ⅱ、Ⅲ组蛋白组成及单克隆抗体反应性更接近欧洲菌株。
In this paper, SDS-PAGE, Western blotting. DotELISA and other molecular epidemiological methods were used to analyze the protein composition and monoclonal antibody reactivity of 21 strains of Borrelia burgdorferi from different regions and different origins in China. The study found that the major proteins in Chinese strains showed a high degree of polymorphism and heterogeneity. There were several different phenotypes of the major outer membrane proteins, OspA, OspB and PC, and showed a close correlation. All strains were positive with the monoclonal antibody H9724 (anti-41KO flagellar antigen). Except for several strains, most Chinese strains did not react with H6831 (anti-OspB monoclonal antibody). Chinese strains and monoclonal antibody H5332 (anti-OspA monoclonal antibody) showed different reactivity, strong, weak and no reaction. According to the protein composition of Chinese strains and monoclonal antibody reactivity can be roughly divided into three groups, of which the first group of strains with the North American representative strain B31 exactly the same protein composition and monoclonal antibody reactivity. The protein composition and the reactivity of the monoclonal antibodies in groups II and III are closer to the European strains.
