目的观察人参皂甙Rg3对肝癌Bel-7402细胞凋亡的影响及其可能机制。方法用不同浓度人参皂甙Rg3作用于肝癌Bel-7402细胞24~72 h后,MTT法检测细胞活力;荧光染色观察细胞核的形态学改变;流式细胞术检测细胞凋亡率;Western blot检测细胞caspase-3、Bcl-2和Bax的表达情况。结果人参皂甙Rg3对Bel-7402细胞的生长有明显的增殖抑制作用,呈浓度和时间依赖性。经荧光染色后可观察到典型的凋亡小体。细胞凋亡率明显增高,Bcl-2蛋白表达量逐渐降低,Caspase-3和Bax蛋白表达量逐渐增加,呈明显的浓度依赖性。结论人参皂甙Rg3能诱导体外培养的人肝癌Bel-7402细胞凋亡;下调Bc1-2和上调Bax表达,活化caspase-3是人参皂甙Rg3诱导Bel-7402细胞凋亡的可能机制之一。
Objective To observe the effect of ginsenoside Rg3 on the apoptosis of hepatocellular carcinoma Bel-7402 cells and its possible mechanism. Methods The cells were treated with different concentrations of ginsenoside Rg3 for 24-72 h. The cell viability was measured by MTT assay. The morphological changes of the cells were observed by fluorescence staining. The apoptosis rate was detected by flow cytometry. The expressions of caspase -3, Bcl-2 and Bax expression. Results Ginsenoside Rg3 had a significant inhibitory effect on the growth of Bel-7402 cells in a concentration- and time-dependent manner. Typical apoptotic bodies were observed after fluorescence staining. The apoptosis rate was significantly increased, the expression of Bcl-2 protein decreased gradually, and the expression of Caspase-3 and Bax protein gradually increased in a concentration-dependent manner. Conclusion Ginsenoside Rg3 can induce the apoptosis of Bel-7402 cells in vitro. Down-regulation of Bc1-2 and up-regulation of Bax expression may activate Gp3 to induce apoptosis of Bel-7402 cells induced by ginsenoside Rg3.
