目的:观察胃乃安新制剂对慢性浅表性胃炎的作用。方法:SD大鼠按随机数字表分为正常组(10只)、模型组、胃乃安新制剂高、中、低剂量(9.10,2.87,0.91 g·kg-1)组和胃乃安胶囊(9.10 g·kg-1)组(每组各15只),予模型组及各给药组大鼠自由饮用0.1%碘代乙酰胺3周构建慢性浅表性胃炎模型的同时,各给药组予相应药物干预,模型组和正常对照组予等量自来水灌胃;3周后取胃组织从肉眼观察、病理切片、透射电镜3个层面评价胃乃安新制剂抗碘代乙酰胺致胃黏膜损伤作用,并检测胃组织的丙二醛(MDA)、超氧化物歧化酶(SOD)水平以探讨其可能的作用机制。结果:正常组胃组织未见明显损伤,未见明显炎细胞浸润;与正常组比较,模型组胃组织肉眼可见明显充血水肿,甚至糜烂,镜下见大量炎细胞浸润(P<0.01);由肉眼、病理评分结果,胃乃安新制剂高剂量组、胃乃安胶囊组均有抗胃黏膜损伤作用(与模型组比较P<0.01),且新制剂抗溃疡效果呈剂量依赖性关系;透射电镜观察结果表明,新制剂可明显逆转主细胞、壁细胞的超微结构损伤性改变;新制剂可降低胃组织MDA含量,提升SOD活性,可明显降低胃黏膜的自由基水平(与模型组比较,P<0.01)。结论:胃乃安新制剂对碘代乙酰胺所致慢性浅表性胃炎有显著抗损伤作用,可能通过清除胃黏膜自由基水平的途径发挥作用。
Objective: To observe the effect of Weinaan on the chronic superficial gastritis. Methods: SD rats were randomly divided into normal group (n = 10), model group, Wei Nai An Xin preparation high, medium and low dose (9.10,2.87,0.91 g · kg-1) (9.10 g · kg -1) group (15 rats in each group). The rats in model group and each administration group were given 0.1% iodoacetamide for 3 weeks to construct chronic superficial gastritis model, The rats in the model group and the normal control group were given the same amount of tap water for gavage. After 3 weeks, the stomach tissue was taken for visual observation, pathological sections and transmission electron microscopy were used to evaluate the effect of Gai Naian An anti-iodoacetamide on stomach Mucosal injury, and detect the levels of malondialdehyde (MDA) and superoxide dismutase (SOD) in gastric tissue to explore its possible mechanism. Results: The gastric mucosa of the normal group showed no obvious injury and no obvious inflammatory cell infiltration. Compared with the normal group, the gastric tissue of the model group showed obvious hyperemia and edema, even erosion, with a large number of inflammatory cell infiltration (P <0.01); The results of the naked eye, the pathological score, the high dose of Weinaanxin Preparation and the Weinaan Capsule group all showed anti-gastric mucosal injury (compared with the model group, P <0.01), and the anti-ulcer effect of the new preparation was in a dose-dependent manner. Electron microscopy results show that the new preparation can obviously reverse the ultrastructural damage of the main cells and parietal cells. The new preparation can reduce the content of MDA in the gastric tissue and increase the activity of SOD, and can significantly reduce the free radical level in the gastric mucosa (compared with the model group , P <0.01). CONCLUSION: Weinaoanxin has a significant anti-injury effect on chronic superficial gastritis caused by Iodoacetamide, which may play a role in clearing the gastric mucosal free radical levels.
