为了探索人白血病骨髓造血微环境的异常所在,本实验采用胶体金标记及银增染的免疫电镜方法,检测了急性白血病患者及正常人骨髓长期培养的基质细胞某些生长因子、粘附分子及细胞外基质的表达情况。结果表明,急性白血病患者长期培养的基质细胞表达的GM-CSF(粒-巨噬系集落刺激因子)较对照组高,淋巴细胞归巢相关粘附分子(HCAM),血管内皮细胞粘附分子-1(VCAM-1)和细胞间粘附分子-1(ICAM-1)较对照组低,纤连蛋白(Fn)和Ⅳ型胶原(Col Ⅳ)与对照组无差别。结论提示急性白血病患者骨髓长期培养的基质细胞某些生长因子和粘附分子表达异常。
In order to explore the abnormalities of the hematopoietic microenvironment of human leukemia bone marrow, certain growth factors and adhesion molecules of stromal cells cultured in acute leukemia patients and normal human bone marrow were detected by immunogold electron microscopy using colloidal gold staining and silver staining. The results showed that the long-term cultured stromal cells in patients with acute leukemia express higher GM-CSF (granulocyte macrophage colony stimulating factor) than the control group, lymphocyte homing-associated adhesion molecule (HCAM), vascular endothelial cell adhesion molecule- 1 (VCAM-1) and intercellular adhesion molecule-1 (ICAM-1) were lower than the control group, and fibronectin (Fn) and type IV collagen (Col IV) were not different from the control group. The results suggest that the expression of certain growth factors and adhesion molecules in stromal cells cultured in long-term bone marrow of patients with acute leukemia is abnormal.
