目的:探讨Exendin-4对高糖诱导的心肌细胞氧化应激的影响。方法:通过酶消化法获取乳鼠心肌细胞,在体外高糖暴露条件下,采用不同浓度的Exendin-4处理,MTT法检测细胞的活性,同时运用生化试剂盒测定上清液中丙二醛(MDA)、超氧化物歧化酶(SOD)、还原型谷胱甘肽(GSH)的水平。结果:与对照组相比,高糖环境下的心肌细胞活性明显减低(P<0.05),MDA含量明显增加(P<0.05),SOD及GSH含量明显减少(P<0.05),而给予Exendin-4预处理后,能逆转上述指标的变化,在浓度为1nmol/L和10nmol/L时具有统计学意义(P<0.05)。结论:高浓度葡萄糖通过氧化应激作用诱导心肌细胞损伤,Exendin-4通过抗氧化作用对心肌细胞起到较好的保护性作用。
Objective: To investigate the effect of Exendin-4 on oxidative stress in cardiomyocytes induced by high glucose. Methods: The neonatal rat cardiomyocytes were obtained by enzymatic digestion. The cells were treated with different concentrations of Exendin-4 under high glucose exposure conditions. The cell viability was measured by MTT assay. Meanwhile, the contents of malondialdehyde MDA), superoxide dismutase (SOD), reduced glutathione (GSH) levels. Results: Compared with the control group, the activity of cardiomyocytes was significantly decreased (P <0.05), the content of MDA was significantly increased (P <0.05) and the content of SOD and GSH was decreased significantly (P <0.05) 4 pretreatment can reverse the above changes in the indicators, at a concentration of 1nmol / L and 10nmol / L was statistically significant (P <0.05). CONCLUSION: High concentration glucose induces cardiomyocyte injury through oxidative stress. Exendin-4 exerts a protective effect on cardiomyocytes through antioxidation.
