目的筛选肝细胞癌(HCC)相关基因,探讨其在HCC发病中的临床意义。方法用差异显示技术对比研究正常肝组织、HCC组织及癌旁肝组织之间mRNA的表达差异,以肝癌cDNA片段MRG98.2为探针,对10例HCC及癌旁肝组织进行斑点印渍分析,并通过逆转录聚合酶链反应(RT-PCR)方法检测这些标本血管内皮生长因子(VEGF)mRNA的表达水平。结果从HCC组织中获得的差示片段MRG98.2在7例HCC标本中表达阳性(70%),癌旁肝组织弱表达(2/10)或无表达。 VEGF mRNA在7例MRG98.2阳性表达的HCC组织中6例表达上调。结论 MRG98.2是HCC相关的基因片段,其表达与VEGF mRNA相关,并可能与肿瘤浸润转移、患者的预后不良有关。
Objective To screen the related genes of hepatocellular carcinoma (HCC) and discuss its clinical significance in the pathogenesis of HCC. Methods Differential display technique was used to compare the expression of mRNA between normal liver tissue, HCC tissue and adjacent liver tissues. The liver cancer cDNA fragment MRG98.2 was used as a probe to perform spot blot analysis on 10 cases of HCC and adjacent liver tissues. The expression level of vascular endothelial growth factor (VEGF) mRNA in these specimens was detected by reverse transcription polymerase chain reaction (RT-PCR). Results The differential segment MRG98.2 obtained from HCC tissue was positive in 7 HCC specimens (70%), and the para-carcinoma tissue was weakly expressed (2/10) or not. VEGF mRNA was up-regulated in 6 of 7 HCC tissues positively expressed by MRG98.2. Conclusion MRG98.2 is a gene fragment associated with HCC. Its expression is related to VEGF mRNA and may be related to tumor invasion and metastasis and poor prognosis of patients.
