目的探讨水通道蛋白7(AQP7)基因多态性及单倍型与肥胖的相关性。方法将1559例流调人群按WC分为肥胖组和非肥胖组(对照组)。应用PCR及直接基因测序方法对AQP7基因SNP-1123和SNP-953位点进行基因分型,并构建单倍型。结果肥胖组和对照组SNP-1123和SNP-953基因型频率差异无统计学意义,而-1123C/T等位基因频率差异有统计学意义(P<0.05)。性别分层后女性-1123CC型与-1123XT(CT+TT)型比较,发生肥胖风险增加(P<0.01,OR=1.48,95%CI:1.12~1.97);两组中女性SNP-1123T-SNP-953A单倍型肥胖风险减少(P<0.01,OR=0.71,95%CI:0.56~0.92)。结论女性中,AQP7基因SNP-1123CC型可能增加患肥胖风险;女性SNP-1123T-SNP-953A单倍型可能减少患肥胖风险。
Objective To investigate the association of aquaporin 7 (AQP7) gene polymorphism and haplotype with obesity. Methods A total of 1559 cases of rheumatoid arthritis were divided into obesity group and non-obese group according to WC (control group). The AQP7 gene SNP-1123 and SNP-953 loci were genotyped by PCR and direct gene sequencing to construct haplotypes. Results The frequencies of SNP-1123 and SNP-953 genotypes in obesity group and control group were not significantly different (P <0.05). However, the frequency of -1123C / T allele was statistically significant (P <0.05). There was an increased risk of obesity in female-123CC type-1123XT (CT + TT) group after sex stratification (P <0.01, OR = 1.48, 95% CI: 1.12-1.97); female SNP-1123T-SNP -953A haplotype obesity risk reduction (P <0.01, OR = 0.71, 95% CI: 0.56 to 0.92). Conclusion Female AQP7 SNP-1123CC may increase the risk of obesity. Female SNP-1123T-SNP-953A haplotype may reduce the risk of obesity.
