目的探讨干扰素诱导的跨膜蛋白-1(IFITM1)基因在肿瘤组织中的表达,血清抗IFITM1的抗体反应以及对临床诊断结直肠癌的意义。方法应用半定量RT-PCR方法检测IFITM1基因mRNA在正常大肠粘膜、结直肠癌组织、大肠炎性息肉、腺瘤性息肉、胃癌、食管癌和肝癌组织中的表达。采用Western印迹法检测患者血清抗IFITM1的抗体反应。分析有IFITM1基因表达癌肿的病理特点。结果结直肠癌组织中IFITM1基因mRNA表达率为47.4%(18/38),显著高于大肠腺瘤性息肉的15%(3/20)和胃癌组织的4.8%(1/21)(P<0.05),而正常大肠粘膜、大肠炎性息肉、食管癌和肝癌组织均无表达。结直肠癌组织IFITM1基因mRNA呈强表达,其表达水平(0.8048±0.2273)显著高于大肠腺瘤性息肉(0.4447±0.0989)(P<0.001)。结直肠癌血清相应的抗体反应率为36.8%(14/38),明显高于胃癌的9.5%(2/21)(P<0.05),而在食管癌、肝癌、大肠炎性息肉和大肠腺瘤性息肉以及健康人血清则未检测出相应的抗体反应。有IFITM1基因表达的结直肠癌多数直径大于5cm,侵及浆膜,有淋巴结和远处转移,多属DukesC期和D期,以高分化腺癌为主。结论IFITM1基因可能在结直肠癌的发生、发展和转移过程中起重要作用,有望成为临床诊断结直肠癌的一个良好标志物。
Objective To investigate the expression of interferon-inducible transmembrane protein-1 (IFITM1) gene in tumor tissue, the antibody response to serum anti-IFITM1 and its clinical significance in the diagnosis of colorectal cancer. Methods The mRNA expression of IFITM1 was detected by semi-quantitative RT-PCR in normal colorectal mucosa, colorectal cancer, colorectal polyps, adenomatous polyps, gastric cancer, esophageal cancer and hepatocellular carcinoma. Antibody responses to anti-IFITM1 in serum were detected by Western blotting. Analysis of pathological features of IFITM1 gene expression in cancer. Results The mRNA expression rate of IFITM1 in colorectal cancer was 47.4% (18/38), which was significantly higher than that in gastric adenocarcinoma (15% (3/20) and 4.8% (1/21)) (P < 0.05), while normal colorectal mucosa, large intestine inflammatory polyp, esophageal cancer and liver cancer tissues were not expressed. The mRNA expression of IFITM1 in colorectal cancer was significantly higher than that in colorectal adenoma (0.4447 ± 0.0989, 0.8048 ± 0.2273, P <0.001). The corresponding antibody response rate of colorectal cancer serum was 36.8% (14/38), which was significantly higher than that of 9.5% (2/21) of gastric cancer (P <0.05), while in esophageal cancer, liver cancer, colorectal polyps and colorectal adenocarcinoma Oncogenic polyps and healthy human serum did not detect the corresponding antibody response. Colorectal cancer with IFITM1 gene expression is more than 5cm in diameter, invasion and serosa, lymph node and distant metastasis, mostly DukesC and D, mainly to well-differentiated adenocarcinoma. Conclusion The IFITM1 gene may play an important role in the carcinogenesis, development and metastasis of colorectal cancer. It may be a good marker for the clinical diagnosis of colorectal cancer.
