采用环磷酰胺制备免疫低下小鼠模型,探索乳香提取物提高免疫低下小鼠胸腺前体T细胞数量、增殖及上调Th1和Th2细胞因子产生的作用机制。采用灌胃给小鼠口服乳香提取物。灌胃后取小鼠外周血和胸腺细胞悬液,用特异性抗体染色并经FACS观测小鼠上述细胞数量变化。采用胸腺细胞培养法检测胸腺基质细胞对于前体T细胞分化成熟的支持作用;采用实时定量PCR方法分析乳香提取物促进Th1细胞分化的可能机制。结果表明,乳香提取物具有明显上调小鼠胸腺内未成熟T细胞数量;促进胸腺基质细胞生长及对T细胞分化成熟的支持作用;T细胞发育相关基因表达格局表明灌胃组小鼠胸腺中细胞因子IL-2、IL-6和IL-1基因表达量较对照组高,转录因子T-bet和gata3表达量也增高。进一步分析表明Notch1和其配体DLL1的基因表达量增加。结果提示乳香提取物上调小鼠Th1/Th2细胞亚群可能是通过介导North1/DLL1信号途径而促进胸腺内T细胞分化与成熟。

The expression pattern of T cell development related genes showed that the expression of T cells in the thymus The gene expression of IL-2, IL-6 and IL-1 was higher than that of the control group, and the expression of T-bet and gata3 also increased. Further analysis showed that Notch1 and its ligand DLL1 gene expression increased. The results suggest that the upregulation of Th1 / Th2 subsets in mouse serum may promote the differentiation and maturation of T cells in the thymus by mediating the North1 / DLL1 signaling pathway.
