目的:观察狼毒大戟根中的不同提取物组分和不同化学成分对耐药型和非耐药型强毒人型结核杆菌的体外抑菌效果。方法:采用不同极性的溶剂对狼毒大戟根进行提取,得到6个提取组分,对其中的醋酸乙酯部分进行了柱层析和制备性高效液相层析分离,分离鉴定出狼毒乙素、狼毒乙素-4-O-β-D-吡喃葡萄糖苷、大黄素甲醚、没食子酸等4个单体成分。采用改良7H10琼脂培养基动态观察6个组分和4个单体成分对耐药型和非耐药型结核杆菌的抑制作用。结果:4种单体成分对耐药型和非耐药型结核杆菌均有显著的抑制作用。其中,狼毒乙素的作用最显著;6个提取组分中。乙醚、醋酸乙酯提取物时非耐药型和耐药型结核杆菌亦有不同程度的抑制作用,醋酸乙酯提取物作用最强。结论:狼毒大戟根中存在着抗结核杆菌特别是抗耐药型结核杆菌的活性部位及有效成分。
OBJECTIVE: To observe the antibacterial effects of different extract components and different chemical components in the root of Scutellaria barbata on resistant and non-resistant virulent human Mycobacterium tuberculosis in vitro. METHODS: Solvent extracts were extracted from the roots of Euphorbia scoparia with different polar solvents. Six extraction components were obtained. The ethyl acetate fraction was separated by column chromatography and preparative high-performance liquid chromatography. Wolf was isolated and identified. Four monomer components, such as toxin B, escin B, 4-O-[beta]-D-glucopyranoside, emodin, and gallic acid. The modified 7H10 agar medium was used to observe the inhibitory effect of 6 components and 4 monomer components on resistant and non-resistant Mycobacterium tuberculosis. RESULTS: The four monomeric components had significant inhibitory effects on drug-resistant and non-resistant strains of Mycobacterium tuberculosis. Among them, the most significant effect of wolftocine was in 6 extracted components. Ethyl acetate and ethyl acetate extracts also had different degrees of inhibitory effects on non-drug resistant and resistant Mycobacterium tuberculosis. Ethyl acetate extract had the strongest effect. Conclusion: There are anti-Mycobacterium tuberculosis in the roots of Euphorbia scoparia, especially the active site and active ingredient of resistant M. tuberculosis.
