目的:探讨胃肠道肿瘤患者外周血Survivin剪接变体△EX3 mRNA的表达对该病的发生及发展和预后的价值。方法:选52例胃肠道肿瘤患者为观察组,35例健康体检人员为对照组。采用PCR技术对其外周血Survivin-△EX3 mRNA表达进行检测。结果:观察组外周血组织中Survivin-△EX3 mRNA相对表达量明显高于对照组[(0.51±0.289)vs.(0.003±0.003),P=0.000];低分化者Survivin-△EX3 mRNA阳性率及Survivin-△EX3mRNA相对表达指数均高于中高分化者(χ2=12.5759,P=0.0004;U=13.0915,P=0.0000);I+II期与III+IV期比较,Survivin-△EX3 mRNA阳性率及Survivin-△EX3 mRNA相对表达指数后者明显比前者升高(χ2=7.0806,P=0.0078;U=13.2359,P=0.0000);淋巴结无转移者与有转移者Survivin-△EX3 mRNA阳性率及Survivin-△EX3 mRNA相对表达指数比较,后者明显高于前者(χ2=8.7970,P=0.0030;U=13.6479,P=0.0000)结论:Survivin-△EX3 mRNA阳性表达与胃肠道肿瘤具有关;其表达的高低在分化程度、临床病理分期和淋巴结转移方面均有意义。
OBJECTIVE: To investigate the expression of △ EX3 mRNA of Survivin splicing variant in peripheral blood of patients with gastrointestinal cancer and its value in the occurrence, development and prognosis of the disease. Methods: Fifty-two patients with gastrointestinal cancer were selected as the observation group and 35 healthy people as the control group. The expression of Survivin- △ EX3 mRNA in peripheral blood was detected by PCR. Results: The relative expression of Survivin-△ EX3 mRNA in the peripheral blood of the observation group was significantly higher than that of the control group [(0.51 ± 0.289) vs. (0.003 ± 0.003), P = 0.000] (P = 0.0004; U = 13.0915, P = 0.0000). The positive rates of Survivin-△ EX3 mRNA in stage I + II and III + IV And Survivin- △ EX3 mRNA relative expression index was significantly higher than the former (χ2 = 7.0806, P = 0.0078; U = 13.2359, P = 0.0000); lymph node metastasis-free Survivin- △ EX3 mRNA positive rate and The expression of Survivin-△ EX3 mRNA was significantly higher than that of the former (χ2 = 8.7970, P = 0.0030; U = 13.6479, P = 0.0000) The expression of the level of differentiation, clinical pathological stage and lymph node metastasis are meaningful.
