以人参四种不同状态的愈伤组织为材料,采用水状片法和碘-碘化钾法和5%香草醛-浓硫酸法等组织化学反应对其进行细胞形态观察,同时测定了四种不同形态的人参愈伤组织中的多糖含量和皂苷含量。结果:通过对四种不同人参愈伤组织的细胞学观察发现只有呈黄绿色,颗粒状,质地松软的Ⅳ型愈伤组织皂苷含量最高且细胞分散度最佳,易于做为人参与胡萝卜原生质体融合培养的起始材料。
Four kinds of callus from different states of ginseng were used as materials. The morphological changes of the cells were observed by histochemical methods, such as aqueous iodine method and iodine-potassium iodide method, and 5% vanillin-concentrated sulfuric acid method. Four different morphologies Ginseng callus tissue polysaccharide content and saponin content. Results: Cytological observation of four different ginseng callus revealed that only type Ⅳ callus with yellowish green, granular and soft texture had the highest content of saponin and the best cell dispersity, which was easy to be used as protoplast fusion between ginseng and carrot The starting material for cultivation.
