伤科健骨汤对大鼠四肢开放性骨折模型中VEGF表达及局部组织学表现的影响

目的:观察在大鼠四肢开放性骨折模型旱期愈合过程中,伤科健骨汤对血清中血管内皮生长因子(VEGF)表达及骨折端局部组织学改变的影响。方法:将SPF级SD大鼠造成开放性股骨骨折模型,术后将造模成功的大鼠随机分为实验组和对照组各18只。实验组造模后第2天即开始以伤科健骨汤浓缩液灌服,对照组仅给予正常饲养。术后第1周、2周、4周,分批处死动物取材行免疫组化染色及图像分析测定.酶联免疫吸附双抗体夹心法(ELISA)测定血清VEGF表达的强度。结果:实验组术后第2周、4周的VEGF浓度与对照组相比,差异均有显著性意义(P<0.05)。局部组织免疫组化染色及图像分析测定显示,实验组VEGF的表达在1、2、4周时均强于对照组,其中在第2周时染色阳性强度最高,第4周阳性强度降低,但仍强于对照组。结论:伤科健骨汤在大鼠四肢骨折愈合早期可正向调高VEGF水平,从而促进骨折断端微血管新生增殖,加快骨折端骨性连接形成,起到促进骨折愈合的作用。 OBJECTIVE: To observe the effects of “Changke Jiangu Decoction” on the expression of vascular endothelial growth factor (VEGF) in serum and the local histological changes at the fracture site in the process of healing during the dry period in rats with open fractures of the extremities. Methods: An open femoral fracture model was made in SD rats of SPF grade. After successful operation, rats were randomly divided into experimental group and control group with 18 rats in each group. On the second day after model establishment, the experimental group began to take GKJF concentrated liquid and the control group received normal feeding only. Immunohistochemical staining and image analysis were used to determine the serum VEGF expression by enzyme-linked immunosorbent assay (ELISA) in the first, second, fourth week after operation. Results: Compared with the control group, the VEGF concentrations in the experimental group at the second and fourth week after operation were significantly different (P <0.05). Local tissue immunohistochemical staining and image analysis showed that the expression of VEGF in the experimental group was stronger than that of the control group at 1, 2, and 4 weeks, with the highest positive staining intensity at week 2 and the lower positive intensity at 4 weeks, but still strong In the control group. CONCLUSION: The Injury of Ketao Jianggu Decoction can increase the VEGF level in the early stage of the fracture healing of the extremities in rats, and promote the proliferation of the microvascular in the fracture site. It can accelerate the formation of bone junction and promote the fracture healing.
