目的研究尼莫地平对脑缺血再灌注大鼠脑组织差异蛋白的影响,进一步揭示尼莫地平的脑保护作用及抗损伤修复机制。方法将12只大鼠随机分为脑缺血再灌注模型组和尼莫地平组(每组6只),分别于造模前3d灌胃给予等量生理盐水和尼莫地平20 mg·kg~(-1),每日1次,d3于手术前1h给药。线栓法致缺血再灌注24h后,分别提取脑蛋白质,2-DE电泳分离蛋白质,应用ImageMaster 2D V2002.1软件比较分析,找出差异蛋白,酶切消化后进行肽指纹图谱分析,数据检索,初步鉴定差异蛋白质。结果与模型组比较,从尼莫地平组找到了25个差异表达蛋白斑点(10个低表达,15个高表达),鉴定其中6个蛋白质。结论尼莫地平对脑缺血再灌注的保护作用是多个蛋白共同参与的结果。
Objective To study the effect of nimodipine on differential proteins in brain tissue of rats with focal cerebral ischemia-reperfusion, further reveal the protective effect of nimodipine and the mechanism of anti-injury repair. Methods Twelve rats were randomly divided into cerebral ischemia-reperfusion model group and nimodipine group (6 rats in each group). The rats were given intragastric administration of normal saline and nimodipine 20 mg · kg ~ (-1) once daily and d3 at 1 hour before surgery. Protein was extracted by 2-DE electrophoresis after brain ischemia and reperfusion for 24 h by thread plug method. The protein was analyzed by ImageMaster 2D V2002.1 software to identify the differentially expressed proteins. Results Compared with the model group, 25 differentially expressed protein spots (10 low expression and 15 high expression) were found from nimodipine group, of which 6 proteins were identified. Conclusion The protective effect of nimodipine on cerebral ischemia-reperfusion is the result of multiple proteins participating together.
