目的对配体CD154在系统性红斑狼疮(SLE)患者B淋巴细胞中刺激转录因子NF-资B核转移的异常进行研究。方法分离正常人对照、SLE患者外周血B淋巴细胞及扁桃体生发中心B淋巴细胞,并以后者为参照,观察与正常人对照相比,SLE外周血B淋巴细胞中CD154和抗CD154抗体对NF-资B核转移或基础活化的影响。结果①正常人外周血B细胞CD154刺激主要诱导NF-资B亚单位p50、p65及c-Rel进入细胞核,而SLE外周血B细胞与扁桃体B细胞相似,刺激前细胞核内功能性p50和c-Rel基础值即高于正常人对照,CD154刺激只进一步诱导p65进入细胞核;②抗CD154抗体可抑制SLE外周血CD154高表达的B细胞核中p65和c-Rel的基础活化值,亦与扁桃体B细胞相似。结论与正常人对照相比,SLE外周血B淋巴细胞中CD154诱导NF-资B核转移存在显著异常,类似生发中心表型。
OBJECTIVE: To investigate the abnormality of ligand CD154 in stimulating NF-B nuclear translocation in B lymphocytes from patients with systemic lupus erythematosus (SLE). Methods The B lymphocytes from peripheral blood of patients with SLE and B lymphocytes of tonsil germinal centers were isolated and compared with the controls. The levels of CD154 and anti-CD154 antibodies in peripheral blood B lymphocytes of SLE patients were compared with those of normal controls. Capital B nuclear transfer or basal activation. Results ① Normal human peripheral blood CD154 stimulated NF-subunit B subunit p50, p65 and c-Rel into the nucleus, and SLE peripheral blood B cells and tonsil B cells similar to the stimulation before the activation of nuclear p50 and c- The basal value of Rel was higher than that of normal human control, CD154 stimulation only further induced p65 to enter the nucleus; ② Anti-CD154 antibody could inhibit basal activation of p65 and c-Rel in CD154 highly expressed B cell nuclei of SLE peripheral blood, similar. Conclusion Compared with normal controls, CD154-induced nuclear translocation of NF-B in SLE peripheral blood lymphocytes is significantly abnormal, similar to the germinal center phenotype.
