以维生素C为阳性对照组,比较了参芪通脉益智方不同提取工艺的体外抗氧化活性,并采用灰关联度分析探讨提取物指纹图谱各特征峰对其体外抗氧化作用贡献大小。结果显示参芪通脉益智方水提法、水提醇沉法、醇提法、醇水双提法所得提取物的IC_(50)依次为0.801 4,0.859 1,0.796 1,0.918 0 g·L~(-1),其中醇提法提取物体外抗氧化活性最强,IC_(50)最小,是抗氧化活性成分的最优提取方法,当药材质量浓度达到一定程度时,自由基清除率与维生素C对照组相近。灰关联度分析得出各成分对抗氧化作用贡献大小顺序(按特征峰编号)4>3>33>53>9>10>11>34>15>59>8>61>52>20>42>18>29。谱效关系的初步探索,为中药复方提取工艺及药效物质基础提供了一定参考。
Vitamin C was used as a positive control group. The antioxidant activities of different extraction processes of Shenqi Tongmai Yizhi Decoction were compared. The gray relational analysis was used to investigate the contribution of each characteristic peak of extracts to their antioxidant activity in vitro. The results showed that the IC 50 of the extract from Shenqi Tongmai Yizhifang water extract, water extraction and alcohol precipitation method, alcohol extraction method and alcohol-water double extraction method were 0.801 4, 0.859 1, 0.766 1 and 0.918 0 g · L ~ (-1). Among them, the extract of alcohol extract has the strongest antioxidant activity and the lowest IC 50 in vitro, which is the optimal extraction method of antioxidant active ingredients. Gray correlation analysis showed that the contribution of each component to the antioxidant effect was in the order of the number of peaks 4> 3> 33> 53> 9> 10> 11> 34> 15> 59> 8> 61> 52> 20> 42> 18> 29. The preliminary exploration of spectrum-efficiency relationship provided some references for the extraction of the traditional Chinese medicine compound and its material basis.
