目的:研究二萜类化合物土荆皮乙酸(pseudolaric acid B)诱导膀胱癌5637细胞凋亡的作用,探讨其作用机制。方法:MTT法测定不同浓度土荆皮乙酸作用后5637细胞的生长情况;流式细胞术检测细胞周期情况;AnnexinV-FITC/PI双染流式细胞术观察土荆皮乙酸作用后细胞的凋亡情况;免疫印迹法检测survivin和caspase-3蛋白表达的变化。结果:土荆皮乙酸能诱导5637细胞凋亡,并明显抑制survivin的表达,同时对caspase-3有上调作用。结论:土荆皮乙酸在体外可抑制5637细胞增殖,诱导其发生凋亡。土荆皮乙酸诱导5637细胞发生凋亡的生物学效应可能与survivin的表达下降,caspase-3的表达上升有关。
Objective: To study the effect of diterpene pseudolaric acid B on apoptosis of bladder cancer 5637 cells and to explore its mechanism. Methods: MTT assay was used to determine the growth of 5637 cells treated with acetic acid at different concentrations. The cell cycle was detected by flow cytometry. The apoptosis of cells was detected by Annexin V-FITC / PI double staining flow cytometry The changes of survivin and caspase-3 protein expression were detected by Western blot. Results: Acetate could induce the apoptosis of 5637 cells and significantly inhibit the expression of survivin, and upregulate the expression of caspase-3. CONCLUSION: Acetoacetate can inhibit the proliferation of 5637 cells in vitro and induce its apoptosis. The biological effects induced by etoposide-induced apoptosis in 5637 cells may be related to the decrease of survivin expression and the increase of caspase-3 expression.
