目的:应用星点设计-效应面法优化地锦草总黄酮结肠定位片的处方。方法:以果胶、L-HPC用量及包衣增重为考察因素,以2,6,9 h地锦草总黄酮中有效成分槲皮素的累积释放度为评价指标,采用Design-expert软件对试验数据进行多元线性模型和二次多项式模型拟合,得出最佳数学模型,绘制效应图和等高线图,通过重叠等高线图得到最优处方并进行验证。结果:二次多项式模型相关系数优于多元线性模型,为最终拟合模型;最佳处方的各时间点累计释放度的实际值和预测值接近,相对偏差绝对值<5%。结论:通过星点设计-效应面法建立的模型预测性良好,可用于对地锦草总黄酮结肠定位片处方的优化。
OBJECTIVE: To optimize the prescription of the total flavonoids colon biopsy tablet of Sophora flavescens by the method of star point design and response surface methodology. Methods: Taking pectin, L-HPC dosage and weight gain of coating as the investigation factors, the cumulative release of quercetin from the total flavonoids of P. diffusa at 2,6,9 h was used as evaluation index. Design-expert software The experimental data were multivariate linear and quadratic polynomial model fitting, the best mathematical model was drawn, the effect map and contour map were drawn, and the optimal prescription was obtained through overlapping contour map and verified. Results: The correlation coefficient of quadratic polynomial model was better than multivariate linear model, which was the final fitting model. The actual value of cumulative release at each time point of the best prescription was close to the predicted value, and the relative deviation was less than 5%. CONCLUSION: The model established by the apical design-response surface method is of good predictability and can be used to optimize the formulation of the total flavone colonium tablet.
