目的研究肺癌干细胞中piR-932的表达,以期为肺癌的治疗提供参考。方法应用piRNA芯片检测肺癌干细胞中piRNAs的表达,应用基因芯片检测肺癌干细胞中基因表达的差异,并检测表达下调的基因之一——Latexin的甲基化程度。结果经诱导至EMT的肺癌干细胞中,piR-932的表达明显增高,而Latexin的表达显著下降,并且Latexin启动子区域CpG岛发生了明显的甲基化。结论 piR-932可能通过促进Latexin的甲基化而正调节肺癌干细胞的EMT过程,它可作为肺癌治疗的一个潜在靶点。
Objective To study the expression of piR-932 in lung cancer stem cells in order to provide a reference for the treatment of lung cancer. Methods piRNAs were used to detect the expression of piRNAs in lung cancer stem cells. The gene expression profiles of lung cancer stem cells were detected by microarray. The methylation level of Latexin, one of the genes that was down-regulated, was detected. Results The expression of piR-932 was significantly increased in lung cancer stem cells induced by EMT, while the expression of Latexin was significantly decreased, and the methylation of CpG island in latexin promoter region was obvious. Conclusion piR-932 may regulate the EMT process of lung cancer stem cells by promoting the methylation of Latexin, which may serve as a potential target for the treatment of lung cancer.
