本文报道用反相高效液相色谱法测定血清和尿液中洛美沙星浓度,固定相用C_(18)柱,流动相为甲醇/0.008M磷酸盐缓冲液/0.5M四丁基溴化铵(25:75:4,V/V/V).荧光检测激发波长280nm,发射波长455nm,以外标法定量。洛美沙星的最低检出限为0.06μg/ml,线性范围为0.125-8.μg/ml.线性关系良好(r=0.9999),日内变异系数2.87%,日间变异系数2.94%,回收率75.8%~80.9%。
In this paper, the concentrations of lomefloxacin in serum and urine were determined by reversed-phase high performance liquid chromatography. The stationary phase was eluted with C 18 column and the mobile phase was methanol / 0.008 M phosphate buffer / 0.5 M tetrabutyl bromide Ammonium chloride (25: 75: 4, V / V / V). Fluorescence detection excitation wavelength 280nm, emission wavelength 455nm, external standard method. The minimum detectable limit of lomefloxacin was 0.06 μg / ml with a linear range of 0.125-8. μg / ml. The linearity was good (r = 0.9999), the intra-day coefficient of variation was 2.87%, the intra-day coefficient of variation was 2.94% and the recovery was 75.8% -80.9%.
