目的建立一种新型基于多肽标准品体系的人乳α-乳白蛋白的定量检测方法。方法从人乳α-乳白蛋白筛选出特异性多肽,并建立对应的检测方法;利用化学方法人工合成特异性多肽、同位素标记特异性多肽和内标。结果本方法所选择的多肽具有高度特异性,α-乳白蛋白的检测灵敏度为8.0 mg/100 g,精密度均小于5.22%,回收率均在97.21%~102.45%之间。结论本方法建立了利用特异性多肽检测人α-乳白蛋白的方法,具有较高的准确度、灵敏度和稳定性。通过对447份人乳样品进行检测,初步探索了人乳中α-乳白蛋白的含量范围,为人乳化配方奶粉的华人配方奠定理论基础。
OBJECTIVE To establish a new method for quantitative detection of human milk alpha-lactalbumin based on peptide standard system. Methods The specific peptides were screened from human milk α-lactalbumin, and the corresponding detection methods were established. The specific polypeptides and isotope-labeled specific peptides and internal standard were synthesized by chemical methods. Results The peptides selected by this method were highly specific. The detection sensitivity of α-lactalbumin was 8.0 mg / 100 g, the precision was less than 5.22% and the recoveries were between 97.21% and 102.45%. Conclusion This method established a method for detecting human α-lactalbumin using specific peptides with high accuracy, sensitivity and stability. By testing 447 samples of human milk, the content of α-lactalbumin in human milk was initially explored, laying a theoretical foundation for Chinese formula of human milk formula.
