目的 :研究胰岛素对心肌成纤维细胞胞外基质纤维连接蛋白 (fibronectin ,FN)、层粘连蛋白 (lami nin ,LN)及细胞因子转化生长因子 β1(TGFβ1)表达的影响 ,探讨其在心肌肥厚中的作用。方法 :乳鼠心肌成纤维细胞的培养并鉴定 ;用Streptavidin Peroxi dase (SP)染色方法 ,比较胰岛素实验组和对照组乳鼠心肌成纤维细胞FN、LN及TGFβ1的表达。结果 :培养的细胞经形态学观察和波形蛋白 (vimentin)SP染色 ,证实为心肌成纤维细胞 ;10 -8mol·L-1胰岛素实验组细胞膜上FN和细胞浆内TGFβ1的表达明显强于对照组 (P <0 .0 1) ;LN在培养的心肌成纤维细胞上基本不表达。结论 :在体外 ,胰岛素具有促进心肌成纤维细胞胞外基质FN堆积的作用 ,可能参与了心肌肥厚的发生和发展 ,其机制可能与胰岛素促进心肌成纤维细胞TGFβ1的合成与分泌有关。
OBJECTIVE: To study the effect of insulin on the expression of extracellular matrix fibronectin (FN), laminin (LN) and TGFβ1 in cardiac fibroblasts, and to explore the role of insulin in myocardial hypertrophy Role. Methods: Fetal heart fibroblasts were cultured and identified. The expression of FN, LN and TGFβ1 in myocardial fibroblasts of experimental and control groups were compared by Streptavidin Peroxiase (SP) staining. RESULTS: The cultured cells were stained by vimentin SP and confirmed by morphological observation and vimentin SP staining. The expression of FN and TGFβ1 in the cytoplasm of 10 -8 mol·L-1 insulin group was significantly stronger than that of the control group (P <0.01). LN was not expressed in cultured cardiac fibroblasts. CONCLUSION: In vitro, insulin has the effect of promoting FN accumulation in extracellular matrix of cardiac fibroblasts, which may be involved in the occurrence and development of cardiac hypertrophy. Its mechanism may be related to the insulin synthesis and secretion of TGFβ1 in cardiac fibroblasts.
