目的对海洋来源放线菌Actinoalloteichus cyanogriseus WH1-2216-6产浅蓝霉素A的发酵条件进行优化,提高浅蓝霉素A的产量。方法对培养基的组成、接种量、起始pH、盐度、发酵时间、前体浓度及大孔吸附树脂添加量等条件的研究,通过单因素和正交试验,选择最优发酵条件;结果获得了浅蓝霉素A的最佳发酵条件,培养基组成(可溶性淀粉20g,甘油20g,蛋白胨20g,CaCO32g,2-吡啶甲酸400mg,XAD-16大孔吸附树脂50g,陈海水1L)、装液量150/500mL(体积分数)、5d种龄、3.3%盐度、起始pH=7.5、摇床(180r·min-1,28℃)发酵12d。结论以最佳发酵条件发酵,优化后的浅蓝霉素A的产量为优化前的7.0倍,达到610.5mg/L。
OBJECTIVE To optimize the fermentation conditions of actinomycetes Actinoplanesichis cyanogriseus WH1-2216-6 producing penicillin A and increase the yield of azapentamycin A. Methods The conditions of medium composition, inoculum size, initial pH, salinity, fermentation time, precursor concentration and macroporous resin addition were studied. The optimal fermentation conditions were selected by single factor and orthogonal test. Results The optimal fermentation conditions were obtained as follows: medium (20g of soluble starch, 20g of glycerol, 20g of peptone, 32g of CaCO, 400mg of 2-picolinic acid, 50g of XAD-16 macroporous adsorption resin and 1L of Chen Haishui) Liquid volume 150 / 500mL (volume fraction), 5d kinds of age, 3.3% salinity, initial pH = 7.5, shaker (180r · min-1,28 ℃) fermentation 12d. Conclusion The optimum fermentation conditions for the fermentation, the optimized production of light blue penicillin A 7.0 times the pre-optimization, to 610.5mg / L.
