DNA的分子组成,对其双螺旋结构的变性有影响,一般GC含量高的DNA不易变性。硷基组成的研究为进一步分析DNA、制备探针、分子杂交等基因工程的研究提供基础。为此,我们对两型卫氏并殖吸虫(3np·w,2np·w)及怡乐村并殖吸虫(p·i)的硷基组成进行了检测。 一、虫体来源与接种 除怡乐村并殖吸虫囊蚴采自辽宁省营口县水源乡的蟛蜞外,其它操作同文献[王恩荣等:中国寄生虫学与寄生虫病杂志1991;9(1):46]。 二、染色体DNA的制备 按Leornard所述方法,提取DNA。经测定OD260nm/OD280nm比值分别为1.65、1.73与1.75;DNA含量介于1.7~2.3μg/μl之间。含量与纯度达实验要求。
