目的探讨脂联素对高浓度葡萄糖诱导的原代大鼠胰岛β细胞凋亡的影响及其可能机制。方法分离和培养原代胰岛细胞,分组干预并检测胰岛细胞凋亡率及相关基因表达水平。结果高糖组胰岛β细胞凋亡率、Caspase-3和Bax mRNA表达量显著高于正常葡萄糖和高糖联合脂联素组(P<0.01)。结论脂联素可以抑制高糖诱导的大鼠胰岛β细胞凋亡,其作用机制可能与凋亡相关基因Caspase-3与Bax基因表达下调有关。
Objective To investigate the effect of adiponectin on the apoptosis of islet β cells in primary rat induced by high glucose and its possible mechanism. Methods Primary islet cells were isolated and cultured. Intervention was performed by grouping, and the apoptosis rate of islet cells and related gene expression were detected. Results The apoptosis rate of pancreatic β cells, the expression of Caspase-3 and Bax mRNA in high glucose group were significantly higher than those in normal glucose group and high glucose group (P <0.01). Conclusion Adiponectin can inhibit high glucose-induced pancreatic β-cell apoptosis in rats. The mechanism may be related to the down-regulation of apoptosis-related genes Caspase-3 and Bax gene expression.
