目的:通过高效液相色谱法荧光测定肝癌耐药细胞内阿霉素的含量。方法:肝癌耐药细胞在培养过程中加入阿霉素处理后,用高效液相色谱法荧光测定细胞内阿霉素的含量。结果:阿霉素在4.0~20.0pg范围内呈现良好的浓度-峰高的线性关系(r=0.999 7),重复性好(RSD=0.49%),最低检出限量低(2.0pg),加药培养后肝癌耐药细胞内均检出阿霉素,各实验组与对照组差异有显著性(P<0.01)。结论:该法稳定、可靠。可用于肿瘤细胞内阿霉素的含量测定及药代动力学研究。
Objective: To determine the content of doxorubicin in drug-resistant hepatocellular carcinoma cells by high performance liquid chromatography (HPLC). Methods: Drug-resistant hepatocarcinoma cells were treated with doxorubicin during the culture process, and intracellular doxorubicin levels were determined by high performance liquid chromatography (HPLC). RESULTS: Doxorubicin showed a good linearity (r = 0.999 7) with good reproducibility (RSD = 0.49%) and low detectable limit (2.0 pg) in the range of 4.0-20.0pg Doxorubicin was detected in drug-resistant hepatocarcinoma cells, and there was a significant difference between the experimental and control groups (P <0.01). Conclusion: The method is stable and reliable. It can be used for the determination of adriamycin in tumor cells and its pharmacokinetics.
