目的研究顺铂作用于卵巢癌CAOV3细胞后,端粒酶活性、细胞凋亡和细胞周期变化与细胞耐药性的关系。方法用顺铂处理卵巢癌CAOV3细胞,端粒酶重复序列扩增(TRAP)-银染法测定端粒酶活性,流式细胞仪观察细胞凋亡和细胞周期的改变。结果顺铂(DDP)处理卵巢癌CAOV3细胞后,CAOV3细胞端粒酶活性受到抑制,细胞周期受阻于G2/M期,未见细胞凋亡峰改变。结论CAOV3细胞对顺铂的耐药性通过多因素起作用,DDP对端粒酶活性具有抑制作用,可以改变细胞周期,但对细胞凋亡无明显影响。
Objective To investigate the relationship between telomerase activity, apoptosis and cell cycle changes and drug resistance in cisplatin-treated ovarian cancer cell line CAOV3. Methods The ovarian cancer cell line CAOV3 was treated with cisplatin. The telomerase activity was determined by telomeric repeat amplification (TRAP) - silver staining. The apoptosis and cell cycle were observed by flow cytometry. Results After treated with cisplatin (DDP), the telomerase activity of CAOV3 cells was inhibited and the cell cycle was arrested in G2 / M phase. The apoptosis peak was not observed. Conclusions The drug resistance of CAOV3 cells to cisplatin has a multifactorial role. DDP can inhibit telomerase activity and change the cell cycle, but it has no obvious effect on apoptosis.
