目的检测环氧化酶-2(Cyclooxygenase-2,COX-2)mRNA在人非小细胞肺癌、癌旁及正常肺组织中的表达情况,探讨COX-2mRNA的表达在非小细胞肺癌发生、发展中的意义。方法应用逆转录-聚合酶链反应(RT-PCR)技术检测20例非小细胞肺癌、癌旁及正常肺组织中COX-2mRNA表达水平。结果COX-2mRNA在非小细胞肺癌组织中的表达水平为(1.412±0.375),高于其在癌旁组织中的(0.595±0.296)和正常肺组织中的(0.477±0.105),差异具有显著性(P<0.01);COX-2mRNA表达与患者性别、年龄及肿瘤大小、生长部位均无显著性相关(P>0.05),而与肿瘤组织学分型、分化程度、临床分期及是否淋巴结转移密切相关(P<0.01或P<0.05)。结论COX-2mRNA表达水平在人非小细胞肺癌组织中增高,并与非小细胞肺癌淋巴结转移、远处转移、临床分期等临床病理特征密切相关,COX-2mRNA表达在非小细胞肺癌发生、发展、转移过程中发挥重要作用。
Objective To detect the expression of Cyclooxygenase-2 (COX-2) mRNA in human non-small cell lung cancer, paracancer and normal lung tissue and to investigate the expression of COX-2 mRNA in non-small cell lung cancer Meaning. Methods Reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the expression of COX-2 mRNA in 20 non-small cell lung cancer tissues and adjacent normal lung tissues. Results The expression level of COX-2 mRNA in non-small cell lung cancer was (1.412 ± 0.375), which was significantly higher than that in non-small cell lung cancer (0.595 ± 0.296) and normal lung tissue (0.477 ± 0.105) (P <0.01). The expression of COX-2 mRNA had no significant correlation with the gender, age, tumor size and growth location (P> 0.05), but closely correlated with tumor histological type, differentiation degree, clinical stage and lymph node metastasis (P <0.01 or P <0.05). Conclusions The expression of COX-2 mRNA is increased in human non-small cell lung cancer and is closely related to the clinicopathological characteristics of lymph node metastasis, distant metastasis and clinical stage in non-small cell lung cancer. The expression of COX-2mRNA in non-small cell lung cancer , Transfer process plays an important role.
