目的以分离纯化的42株野生大豆内生真菌为材料,分析内生真菌菌丝及发酵液中次生代谢产物的积累,筛选高产次生代谢产物内生菌株。方法采用液体发酵摇瓶培养法及紫外分光光度法对不同内生真菌菌丝及发酵液中的总三萜、黄酮、多糖、多酚与总酚的量进行检测。结果 42株野生大豆内生真菌菌丝中,筛选获得高质量分数多糖菌株为Y2S9、WDL3和WDL2,质量分数最高可达85.51 mg/g;高质量分数黄酮菌株为Y1S9和Y6S8,质量分数分别为2.99和2.24 mg/g;高质量分数三萜菌株为Y6S8、Y2R1、Y6R2,质量分数分别为25.55、17.89、15.96 mg/g;而WDS7总酚量最高,为22.65 mg/g。内生真菌发酵液中,黄酮量较高菌株为Y1S8和SYS4,其质量浓度分别为3.11和4.81 mg/L;总三萜质量浓度较高的菌株为YSYL2、Y1S8、WDL3,质量浓度分别为17.46、35.00、42.98 mg/L。结论不同生境可能是导致次生代谢产物量差异的主要原因,本研究筛选出高产次生代谢产物野生大豆内生真菌菌株11株,为野生大豆内生真菌功能挖掘及新药筛选奠定了基础。
OBJECTIVE To analyze the accumulation of secondary metabolites in endophytic fungi mycelium and fermentation broth of 42 wild soybean endophytic fungi isolated and purified, and to screen endophytic strains with high yield of secondary metabolites. Methods The total triterpenoids, flavonoids, polysaccharides, polyphenols and total phenols in different endophytic fungi mycelium and fermentation broth were detected by liquid fermentation shake flask culture and UV spectrophotometry. Results Among 42 wild soybean endophytic fungi mycelia, the high quality fraction polysaccharides were screened as Y2S9, WDL3 and WDL2 with the highest concentration of 85.51 mg / g, the high quality fraction of Y1S9 and Y6S8, the mass fractions were 2.99 and 2.24 mg / g, respectively. The high-quality fraction triterpenes were Y6S8, Y2R1 and Y6R2 with the mass fractions of 25.55, 17.89 and 15.96 mg / g, respectively, while WDS7 had the highest total phenol concentration of 22.65 mg / g. The endophytic fungi fermentation broth, the higher flavonoids strains Y1S8 and SYS4, the mass concentrations were 3.11 and 4.81 mg / L; total triterpenoids higher concentration strains YSYL2, Y1S8, WDL3, the mass concentrations were 17.46 , 35.00, 42.98 mg / L. Conclusion Different habitats may be the main reason for the differences in secondary metabolites. In this study, 11 strains of endophytic fungi producing wild soybean with high yield were screened, which laid the foundation for the functional excision of wild endophytic fungi and the screening of new drugs.
