目的研究小鼠淋巴瘤细胞L5178/Tk+/-3.7.2CTk位点杂合性,探讨Tk+/-基因稳定性,为实验检测过程中的质量控制提供可行的技术指标和实验依据。方法PCR分析L5178YTk+/-3.7.2C小鼠淋巴瘤细胞Tk位点的扩增产物。结果扩增产物为单一带型,未发现细胞株Tk杂合性丢失。结论被测试细胞株没有发生基因内和基因间突变、缺失、重组和移位等事件,说明细胞的遗传特性是稳定的。
Objective To investigate the heterozygosity of L5178 / Tk +/- 3.7.2CTk in mouse lymphoma cells and to investigate the stability of Tk +/- gene, and to provide feasible technical and experimental evidences for the quality control during the test. Methods The amplification products of Tk sites in L5178YTk +/- 3.7.2C mouse lymphoma cells were analyzed by PCR. Results The amplified product was single band and no loss of Tk heterozygosity was found in the cell line. CONCLUSION: The in vitro and in vivo genetic mutations, deletions, recombination and translocation of the tested cell lines have not occurred, indicating that the genetic characteristics of the cells are stable.
