利用响应面优化法,分别优化纯水和水醇混合(V_水∶V_(乙醇)=1∶1)提取大海马活性成分的最佳工艺条件。在单因素试验基础上,以提取温度、提取时间、液料比为自变量,蛋白质含量和抗氧化能力(DPPH清除率)为响应值,做三因素三水平的响应面回归分析。通过分析各因素的显著性和交互作用,优化得到纯水提取大海马活性成分的最佳工艺条件:提取温度70℃、提取时间4.2 h、液料比207 mL/g。该条件下蛋白质含量为31.89 mg/g,抗氧化能力为31.83%;水醇混合提取的最佳工艺条件:提取温度54℃、提取时间5.2 h、液料比230 m L/g。该条件下蛋白质含量为5.11 mg/g,抗氧化能力为39.76%。
Response surface optimization method was used to optimize the extraction of the active ingredients of the hippocampus from pure water and hydroalcoholic mixture (V_water: V_ (ethanol) = 1: 1). On the basis of single factor experiment, response surface regression analysis of three factors and three levels was carried out based on the extraction temperature, extraction time, ratio of liquid to material as independent variable, protein content and antioxidant capacity (DPPH clearance). By analyzing the significance and interaction of various factors, the optimal conditions for extracting active ingredients of hippocampus from pure water were optimized: the extraction temperature was 70 ℃, the extraction time was 4.2 h and the ratio of liquid to solid was 207 mL / g. Under these conditions, the protein content was 31.89 mg / g and the antioxidant capacity was 31.83%. The optimal conditions for the extraction of hydroalcoholic extract were as follows: the extraction temperature was 54 ℃, the extraction time was 5.2 h and the liquid-to-liquid ratio was 230 m L / g. Under these conditions, the protein content was 5.11 mg / g and the antioxidant capacity was 39.76%.
