对北京十三陵苗圃北京蟒山、山西孝义和甘肃两当3个种源的8年生白皮松1年生针叶进行了电阻抗图谱(EIS)分析,测定各种源不经冷冻处理针叶的EIS参数,以及超氧化物歧化酶(SOD)活性、过氧化物酶(POD)活性、丙二醛(MDA)含量、可溶性蛋白质、脯氨酸、叶绿素和干物质含量等生理指标的变化;分析EIS参数和生理指标之间的相关性;用褐变法测定了针叶的抗寒性。结果表明:(1)针叶的EIS参数和生理指标随着抗寒性的提高发生变化,不同种源的EIS参数和某些生理指标在不同抗寒锻炼阶段,尤其在抗寒锻炼初期存在显著差异,并与不同种源抗寒性差异吻合;(2)EIS参数细胞膜时间恒量τm和胞外电阻re与针叶的叶绿素含量存在极显著负相关(r分别为-0.91和-0.68),β与针叶的干物质含量存在极显著负相关(r=-0.74),而胞内电阻ri和re与针叶的干物质含量存在极显著正相关(r分别为0.69和0.58),β与MDA含量存在极显著正相关(r=0.68);(3)β与褐变法求得的抗寒性存在极显著正相关(r=0.85),ri和re存在极显著负相关(r分别为-0.79和-0.66)。
The electrical impedance spectroscopy (EIS) analysis of one-year-old coniferous perennial coniferous white peonies with three provenances, Beijing Mangshan, Xiaoyi and Gansu in Ming Tombs Nursery in Beijing, EIS parameters and the changes of physiological indexes such as superoxide dismutase (SOD) activity, peroxidase (POD) activity, malondialdehyde (MDA) content, soluble protein, proline, chlorophyll and dry matter content were analyzed. EIS parameters and physiological indicators of the correlation between browning method was used to determine the cold resistance of needles. The results showed that: (1) EIS parameters and physiological indices of needles changed with the increase of cold resistance. EIS parameters and some physiological indexes of different provenances were significantly different in different cold resistance stages, especially in the early stage of cold resistance training (2) The time constant τm and the extracellular resistance re of the EIS parameters were significantly and negatively correlated with the chlorophyll content (r = -0.91 and -0.68, respectively) and β (R = -0.74), but the intracellular resistances ri and re were positively correlated with the dry matter content of needle (r = 0.69 and 0.58, respectively) (R = 0.68). (3) There was a significant positive correlation between β and browning resistance (r = 0.85). There was a significant negative correlation between ri and re (r = -0.79 And -0.66).
