目的探讨Toll样受体4(Tolllikereceptor4,TLR4)在外周血白细胞上的表达及其表达差异在动脉粥样硬化中的意义。方法采用流式细胞术检测50例冠状动脉粥样硬化性心脏病患者及40名健康对照者外周血淋巴细胞、单核细胞和中性粒细胞上TLR4的表达;全自动生化分析仪测定所有入选者血脂水平。结果TLR4在外周血淋巴细胞和中性粒细胞的阳性率平均值低于5%;单核细胞的TLR4阳性率在冠心病组为(55.11±16.30)%,显著高于健康对照组的阳性率(42.91±15.70)%(P<0.01);而冠心病血脂正常组和血脂异常组的TLR4阳性率无统计学意义[(54.16±14.60)%;(56.41±18.30)%,P>0.05]。结论TLR4主要表达于外周血单核细胞,在动脉粥样硬化患者中其阳性率升高;而且TLR4的阳性率高低与患者血脂正常与否无关;TLR4及其介导的天然免疫应答与动脉粥样硬化的发生发展存在一定的相关性。
Objective To investigate the expression of Toll-like receptor 4 (TLR4) on peripheral leukocytes and its significance in atherosclerosis. Methods Flow cytometry was used to detect the expression of TLR4 on peripheral blood lymphocytes, monocytes and neutrophils in 50 patients with coronary atherosclerotic heart disease and 40 healthy controls, and all the patients were selected by automatic biochemical analyzer Blood lipid levels. Results The positive rate of TLR4 in peripheral blood lymphocytes and neutrophils was lower than 5%. The positive rate of TLR4 in monocytes was (55.11 ± 16.30)% in coronary heart disease group, which was significantly higher than that in healthy controls (42.91 ± 15.70)%, respectively (P <0.01). However, the positive rate of TLR4 in patients with coronary heart disease was significantly higher than that in patients with dyslipidemia (54.16 ± 14.60% vs 56.41 ± 18.30%, P> 0.05). Conclusion TLR4 is mainly expressed in peripheral blood mononuclear cells, and the positive rate of TLR4 is elevated in atherosclerotic patients. Moreover, the positive rate of TLR4 is not related to the normal blood lipids in patients. TLR4 and its innate immune response are associated with atherosclerosis There is a certain correlation between the occurrence and development of so-called sclerosis.
