目的深入分析AKR/J小鼠的与胆固醇代谢相关的表型特征及其内在联系,探讨其作为人类疾病模型的价值。方法 AKR/J小鼠为实验组,BABL/C小鼠为对照组,取两组小鼠毛发镜下观察;取两组小鼠的肾上腺、卵巢、睾丸,冷冻切片后进行油红O染色;检测并比较两组小鼠肾上腺相关激素(皮质素、醛固酮、ACTH)水平;检测并比较血常规、血糖、血脂(甘油三酯,总胆固醇)、血电解质及血压。结果 AKR/J小鼠内部毛发结构排列紊乱,肾上腺脂类含量明显少于对照鼠,皮质素、醛固酮、总胆固醇浓度比对照小鼠有显著性降低(P<0.01);血液Na+、Cl-、Mg2+浓度及平均血红蛋白含量低于对照鼠,Ca2+浓度高于对照鼠(p<0.05),其他各项指标均无显著性差别。结论 AKR/J小鼠是人类毛发结构缺陷、肾上腺皮质机能减退等疾病的天然模型,也是胆结石及动脉粥样硬化的抗性模型。
OBJECTIVE: To investigate in depth the phenotypic characteristics of AKR / J mice associated with cholesterol metabolism and its intrinsic relationship, and to explore its value as a human disease model. Methods AKR / J mice were used as experimental group and BABL / C mice were used as control group. The mice in two groups were observed under the microscope by hair; Adrenal gland, ovary, testis and frozen sections were taken for oil red O staining. The levels of adrenal related hormones (corticosterone, aldosterone and ACTH) in the two groups were detected and compared. Blood routine test, blood glucose test, blood lipid (triglyceride, total cholesterol), blood electrolytes and blood pressure were detected and compared. Results The hair structure of AKR / J mice was disordered and the content of adrenal lipid was significantly lower than that of control mice (P <0.01). The concentrations of Na +, Cl-, Mg2 + concentration and mean hemoglobin content were lower than those of control mice, Ca2 + concentration was higher than that of control mice (p <0.05). There was no significant difference between other indexes. Conclusion AKR / J mouse is a natural model of human hair structural defects, adrenal cortical dysfunction and other diseases, but also a model of gallstone and atherosclerosis resistance.
