目的 :探讨化瘀补肾汤对再生障碍性贫血 (简称“再障”)小鼠骨髓微环境及血清造血负调控因子白细胞介素 2 (IL 2 )、γ干扰素 (IFNγ)的影响。方法 :复制免疫介导的再障小鼠模型 ,胃饲中药复方。第 1 1d ,观察骨髓切片组织形态学变化 ,测定骨髓基质细胞体外培养计数及IL 2、IFNγ水平。结果 :再障模型组骨髓增生极度低下 ,骨髓微环境明显受损 ,血清IL 2、IFNγ水平明显增高。而中药组骨髓增生好转 ,骨髓微环境损伤减轻 ,血清IL 2、IFNγ水平较再障组减低。结论 :化瘀补肾汤能促进再障小鼠骨髓微环境的修复 ,减少造血负调控因子的释放 ,从而促进造血功能恢复。
Objective: To explore the effect of Huayu Bushen Decoction on the bone marrow microenvironment and serum hematopoietic negative regulators interleukin 2 (IL 2) and interferon-gamma (IFNγ) in mice with aplastic anemia (abbreviated as “ABC”). Methods: Copy immune-mediated mice model of aplastic anemia, stomach feeding Chinese herbal compound. On the 11th day, the histomorphological changes of bone marrow slices were observed. The in vitro culture counts of bone marrow stromal cells and the levels of IL-2 and IFNγ were measured. Results: The bone marrow proliferation was extremely low in the model group and the bone marrow microenvironment was significantly impaired. The levels of serum IL-2 and IFNγ were significantly increased. The Chinese medicine group improved myeloproliferation, bone marrow microenvironmental injury was reduced, serum IL2, IFNγ levels decreased compared with the aplastic anemia group. Conclusion: Huatan Bushen Decoction can promote the repair of bone marrow microenvironment in aplastic anemia mice, reduce the release of negative hematopoietic regulators, and promote the recovery of hematopoietic function.
