目的建立高效液相色谱法测定中医滋补膏方中橙皮苷含量的方法。方法采用Hypersil ODS2色谱柱(250 mm×4.6 mm,5 m);乙腈-甲酸缓冲液(取甲酸1.7 mL,加水,再加三乙胺1.8 mL,定容至1 000 mL)(20∶80)为流动相;流速为1.0 mL.min 1;检测波长为283 nm,柱温为30℃,进样量为20 L。结果橙皮苷2.1~12.6 g.mL 1内,峰面积与浓度呈良好的线性关系(r=0.999 8),平均回收率和RSD分别为99.1%和1.68%。结论方法简便、可靠,重复性好,可用于中医滋补膏方中橙皮苷含量测定。
Objective To establish a method for the determination of hesperidin in traditional Chinese medicine nourishing cream by high performance liquid chromatography. Methods Hypersil ODS2 column (250 mm × 4.6 mm, 5 m) was used. Acetonitrile-formic acid buffer solution (1.7 mL of formic acid and water, then triethylamine 1.8 mL and volume to 1000 mL) As mobile phase. The flow rate was 1.0 mL.min 1. The detection wavelength was 283 nm and the column temperature was 30 ℃. The injection volume was 20 L. Results There was a good linear relationship between the peak area and the concentration of hesperidin (r = 0.999 8) in the range of 2.1 ~ 12.6 g.mL 1. The average recoveries and RSDs were 99.1% and 1.68%, respectively. Conclusion The method is simple, reliable and reproducible. It can be used for the determination of hesperidin in traditional Chinese medicine nourishing cream.
