目的:观察当归多糖对X射线辐射SD大鼠肾脏氧化应激损伤防治作用及其相关机制。方法:48只雄性大鼠随机分为空白对照组、模型组、贞芪扶正颗粒组、当归多糖低、中、高剂量组,共6组,每组8只,贞芪扶正颗粒组灌喂2.62g/kg贞芪扶正颗粒溶液,当归多糖低、中、高剂量组分别灌喂63.6 mg/kg、127.1 mg/kg和254.2 mg/kg当归多糖,空白对照组和模型组分别灌胃等体积的生理盐水,连续灌胃7d后,用X射线6Gy分两次照射,照射后继续灌胃3天。检测各组小鼠体重、肾脏指数变化情况;肾脏HE病理切片;Cr、BUN肾功能结果;GSH-Px、LPO、MDA、SOD氧化应激功能指标和Nrf2蛋白表达情况。结果:与空白对照组相比,模型组大鼠一般状况差,体重下降,肾脏指数升高;Cr和BUN表达升高;LPO和MDA水平增高,SOD、GSH-Px水平降低;Nrf2蛋白表达升高。与模型组相比,63.6 mg/kg、127.1 mg/kg和254.2 mg/kg当归多糖各组大鼠一般状况尚可,体重增加,肾脏指数降低;Cr和BUN表达下降;LPO和MDA表达降低,SOD、GSH-Px水平升高;Nrf2蛋白表达降低。结论:当归多糖对X射线辐射SD大鼠所致的肾脏氧化应激损伤具有防护作用,可能和Nrf蛋白表达有关。
Objective: To observe the role of Angelica polysaccharide in the prevention and treatment of oxidative stress in kidneys of X-ray-irradiated SD rats and its related mechanisms. Methods: Forty-eight male rats were randomly divided into blank control group, model group, Zhenqi Fuzheng Granule Group, Angelica sinensis polysaccharide low, medium and high dose group, 6 rats in each group, Zhenqi Fuzheng granule group fed 2.62 g / kg Zhenqi Fuzheng granule solution, Angelica polysaccharide low, medium and high dose groups were fed 63.6 mg / kg, 127.1 mg / kg and 254.2 mg / kg Angelica sinensis polysaccharide, respectively, the blank control group and the model group were intragastrically gavage Physiological saline, continuous gavage 7d, with X-ray 6Gy two irradiation, continue to gavage after irradiation for 3 days. The changes of body weight and index of kidney in each group were observed. The pathological changes of renal HE, the renal function of Cr and BUN, the indexes of oxidative stress of GSH-Px, LPO, MDA and SOD and the expression of Nrf2 protein were observed. Results: Compared with the blank control group, the rats in the model group were generally poor in body weight, decreased in kidney weight, increased in kidney index, elevated in Cr and BUN, increased in LPO and MDA, decreased in SOD and GSH-Px, while increased in Nrf2 high. Compared with the model group, the general condition of rats in 63.6 mg / kg, 127.1 mg / kg and 254.2 mg / kg Angelica Sinensis polysaccharides was acceptable, the body weight was increased, the renal index was decreased, the expressions of Cr and BUN were decreased, the expressions of LPO and MDA were decreased, SOD, GSH-Px levels increased; Nrf2 protein expression decreased. Conclusion: Angelica polysaccharide has a protective effect on kidney oxidative stress induced by X-ray irradiation in SD rats, which may be related to the expression of Nrf protein.
