目的:建立高效液相色谱法测定罗布麻叶中芦丁和槲皮素的含量。方法:采用krom asil C18柱(4.6mm×250 mm,5μm);流动相为甲醇(A)-0.08%磷酸(B),梯度洗脱;流速1.0 mL/m in;检测波长360nm。结果:芦丁进样量在0.42~7.10μg范围内(r=0.9998),槲皮素进样量在0.011~0.19μg范围内(r=0.9997)线性关系良好,平均回收率芦丁为98.77%(RSD%=1.2),槲皮素为99.19%(RSD%=1.8)。结论:该方法操作简便,准确,重复性好,可同时测定罗布麻叶中芦丁和槲皮素的含量。
Objective: To establish a HPLC method for the determination of rutin and quercetin in leaves of A. venetum. Methods: A Krom asil C18 column (4.6 mm×250 mm, 5 μm) was used. The mobile phase consisted of methanol (A)-0.08% phosphoric acid (B) with a gradient elution. The flow rate was 1.0 mL/min. The detection wavelength was 360 nm. Results: The rutin injection volume ranged from 0.42 to 7.10 μg (r=0.9998). The quercetin injection volume ranged from 0.011 to 0.19 μg (r=0.9997). The linear relationship was good and the average recovery rate was 98.77%. (RSD%=1.2), quercetin was 99.19% (RSD%=1.8). Conclusion: This method is simple, accurate, and reproducible. It can simultaneously determine the contents of rutin and quercetin in leaves of A. venetum.
