以野生型2型人腺伴随病毒(AAV-2)基因组载体pSSV9-int-为基础,构建了一种携带和表达真核基因的基础AAV载体pSSV9/MulV-neosv,表达了neo基因,并采用该载体组建了携带人白细胞介素-2cDNA的重组AAV载体pSSV9/MulV-neosv-IL-2,转染A549细胞后表达了人白细胞介素-2基因。为应用腺伴随病毒作基因治疗研究提供了实验基础。
Based on the pSSV9-int- vector of the wild-type human adeno-associated virus type 2 (AAV-2) genome, a basic AAV vector pSSV9 / MulV-neosv carrying and expressing a eukaryotic gene was constructed and neo gene was expressed. The recombinant AAV vector pSSV9 / MulV-neosv-IL-2 containing human interleukin-2 cDNA was constructed and transfected into A549 cells to express human interleukin-2 gene. It provides experimental basis for the application of adeno-associated virus in gene therapy research.
