目的探讨选择性COX-2抑制剂Celecoxib对非小细胞肺癌细胞株A549增殖与凋亡方面的作用。方法对非小细胞肺癌细胞株A549使用MTT法测定细胞生长抑制率;Hoechest33258荧光染色观察细胞凋亡;流式细胞仪分析细胞凋亡及细胞周期分布。结果Celecoxib对抑制A549细胞增殖的IC50值为14.7μmol/l;Celecoxib对A549细胞的毒性较大程度上是通过诱导NSCLC细胞的凋亡,Celecoxib5-40μmol/l作用12-48hr的凋亡比率为2.1 ̄44.3%。结论COX-2抑制剂Celecoxib在体外能诱导非小细胞肺癌细胞发生凋亡,抑制细胞增殖。
Objective To investigate the effect of selective COX-2 inhibitor Celecoxib on the proliferation and apoptosis of non-small cell lung cancer cell line A549. Methods The cell growth inhibition rate of non-small cell lung cancer cell line A549 was measured by MTT assay; Hoechst 33258 fluorescence staining was used to observe cell apoptosis; Cell apoptosis and cell cycle distribution were analyzed by flow cytometry. Results The IC50 value of Celecoxib inhibited the proliferation of A549 cells was 14.7μmol/l. The toxicity of Celecoxib to A549 cells was largely induced by apoptosis of NSCLC cells. The apoptosis rate of Celecoxib at 5-40μmol/l was 12-48hr. ~ 44.3%. Conclusion COX-2 inhibitor Celecoxib can induce apoptosis and inhibit cell proliferation in non-small cell lung cancer cells in vitro.
