目的探讨华蟾素(cinobufagin)对人骨肉瘤细胞株MG-63的促凋亡作用。方法采用MTT比色法、透射电镜观察、原位末端标记tunnel法、流式细胞仪检测等方法,观察和检测华蟾素对MG-63的促凋亡作用。结果华蟾素对MG-63骨肉瘤促凋亡作用明显,且呈现时间和浓度依赖性。华蟾素将MG-63细胞阻断在S期,可诱导骨肉瘤细胞凋亡。与甲氨蝶呤对照组相比,不同浓度下华蟾素对MG-63细胞促凋亡作用更显著。结论华蟾素对MG-63骨肉瘤促凋亡明显。华蟾素可用于骨肉瘤治疗。
Objective To investigate the effect of cinobufagin on the apoptosis of human osteosarcoma cell line MG-63. Methods MTT colorimetric method, transmission electron microscopy, in situ end-labeled tunnel method and flow cytometry were used to detect the apoptosis of MG-63 cells induced by cinobufacini. Results Cinobufacini significantly promoted the apoptosis of MG-63 osteosarcoma in a time-and concentration-dependent manner. Cinobufacini blocked MG-63 cells in S phase and induced osteosarcoma cell apoptosis. Compared with methotrexate control group, cinobufacini significantly promoted the apoptosis of MG-63 cells at different concentrations. Conclusion Cinobufacini can promote the apoptosis of MG-63 osteosarcoma. Cinobufacini can be used for osteosarcoma treatment.
