中药健胃愈疡颗粒剂治疗大鼠实验性胃溃疡

目的:研究健胃愈疡颗粒治疗消化性溃疡的机制.方法:采用改良Okabe法复制大鼠胃溃疡模型.76只SD大鼠随机分为4组,除正常组外,其余三组均手术造模,每日灌胃分别饲以蒸馏水和不同药物,连续7d或14d后取溃疡胃组织,作常规病理检查,观察溃疡情况,TUNEL法检测细胞凋亡,免疫组化法检测各指标蛋白表达,原位杂交法法检测各指标mRNA表达.结果:JWYY治疗组在7d和14d后,溃疡指数(1.7±0.5,0.8±0.4)、凋亡指数(3.0±0.4,2.0±0.5)明显降低;MK蛋白(33.7±3.7,38.0±4.1)和mRNA(22.6±2.9,30.5±3.7)、EGFR蛋白(39.6±4.9,31.9±6.8)和mRNA(26.6±5.0,17.4±1.6)表达明显升高,较模型组和胃苏组有显著统计学差别(P<0.01或0.05).JWYY组7d时TNF-α蛋白(18.7±3.3)和mRNA(11.4±3.0)表达显著低于模型组(26.1±3.8,;17.1±2.7;P<0.01),而高于正常组(4.0±0.9,3.3±1.0,P<0.01);14d时各组则无明显统计学差别(P>0.05).JWYY组7d和14d时Bcl-2蛋白(33.5±6.8,32.9±9.6)、mRNA(26.1±4.5,23.0±6.2)及mRNABcl-2/Bax(1.4±0.3,1.5±0.4),蛋白Bcl-2/Bax(1.3±0.3,1.7±0.7)均显著高于模型组(P均<0.05或0.01).结论:JWYY颗粒通过提高Bcl-2、Bcl-2/Bax抑制细胞凋亡,降低TNF-α表达,提高MK、EGFR表达而促进溃疡愈合,提高溃疡愈合质量. Objective: To study the mechanism of Jianweiyuyang granules in the treatment of peptic ulcer. Methods: Rat gastric ulcer model was modified by the modified Okabe method. Seventy-six Sprague-Dawley rats were randomly divided into 4 groups. Except the normal group, the other three groups were operated. The rats were fed with distilled water and different drugs daily for 7 days or 14 days, and the gastric tissues were taken for routine pathological examination to observe the ulcer. TUNEL was used to detect cell apoptosis. Immunohistochemistry was used to detect the expression of various indicators. In situ hybridization was used to detect the mRNA expression of each index. Results: After 7 days and 14 days, the ulcer index (1.7±0.5, 0.8±0.4) and apoptotic index (3.0±0.4, 2.0±0.5) were significantly decreased in the JWYY treatment group; The expression of protein (33.7±3.7, 38.0±4.1) and mRNA (22.6±2.9, 30.5±3.7), EGFR protein (39.6±4.9, 31.9±6.8) and mRNA (26.6±5.0, 17.4±1.6) were significantly higher. There was a significant difference between the model group and Weisu group (P<0.01 or 0.05). The expression of TNF-α protein (18.7±3.3) and mRNA (11.4±3.0) was significantly lower in the JWYY group than in the model group (26.1±3.8) at 7d. 17.1 ± 2.7; P <0.01), but higher than the normal group (4.0 ± 0.9, 3.3 ± 1.0, P <0.01); 14d, there was no significant difference between the groups (P> 0.05). JWYY group 7d and 14d Bcl-2 protein (33.5±6.8, 32.9±9.6) mRNA (26.1±4.5, 23.0±6.2), mRNA Bcl-2/Bax (1.4±0.3, 1.5±0.4) and protein Bcl-2/Bax (1.3±0.3, 1.7±0.7) were significantly higher than those in the model group (P <0.05 or 0.01). Conclusion: JWYY granules can inhibit apoptosis, decrease TNF-α expression, increase the expression of MK and EGFR, promote ulcer healing and improve the quality of ulcer healing by increasing Bcl-2 and Bcl-2/Bax.
