目的采用RP-HPLC法测定小儿腹泻宁中葛根素和大豆苷元含量。方法采用Venusil XBP-C18色谱柱(4.6×250mm,5μm);流动相:甲醇:水(v/v,25:75);柱温:35℃;流速:1.0ml/min;检测波长:250nm。结果葛根素质量浓度在2.0~50.0mg/L内,大豆苷元质量浓度在1.0~25.0mg/L内与峰面积呈良好的线性关系,平均回收率分别为100.02%(RSD0.35%)、97.93%(RSD 1.54%);进样精密度RSD分别为0.40%、1.21%(n=6),方法精密度RSD分别为0.70%、0.91%(n=6)。结论论本方法简便、快速、准确,重复性好,可用于小儿腹泻宁中葛根素和大豆苷元的含量测定。
Objective To determine the contents of puerarin and daidzein in children with diarrhea through RP-HPLC. The method was performed on a Venusil XBP-C18 column (4.6 × 250 mm, 5 μm). The mobile phase was methanol: water (v / v, 25:75). The column temperature was 35 ℃. The flow rate was 1.0 ml / min. Results The mass concentration of puerarin was 2.0 ~ 50.0 mg / L, and the concentration of daidzein was in the range of 1.0 ~ 25.0 mg / L. The average recoveries were 100.02% (RSD 0.35%), The precision of RSD was 0.40% and 1.21% respectively (n = 6). The RSD of precision was 0.70% and 0.91% respectively (n = 6). Conclusion This method is simple, rapid, accurate, reproducible, and can be used for determination of puerarin and daidzein in children with diarrhea.
