目的研究血小板第 4因子(PF4)对60Co γ射线辐射损伤小鼠骨髓的造血防护作用及其机制。方法小鼠腹腔注射PF4后间隔 6 h第 2次注射; 20 h后,给予 7. 5Gy的60Co γ射线照射 1次(全身照射),观察第8天骨髓细胞DNA含量的变化。结果照射后8d,PF4处理组小鼠的骨髓有核细胞中DNA含量及细胞周期各时相,与照射组相比较有明显差异(P<0.01),而与对照组基本相同(P>0.05)。结论PF4能保护小鼠骨髓造血干细胞免受电离辐射引起的损伤。
Objective To study the hematopoietic protective effect of platelet factor 4 (PF4) on mice bone marrow irradiated by 60Co γ-ray and its mechanism. Methods Mice were intraperitoneally injected with PF4 for 6 hours after the second injection; 20 hours later, given 7. The 5Gy 60Co γ-rays were irradiated once (for whole body irradiation), and the changes in DNA content of bone marrow cells on the 8th day were observed. Results 8 days after irradiation, the DNA content and cell cycle of bone marrow nucleated cells in the PF4 treated group were significantly different from those in the irradiated group (P<0.01), but were similar to those in the control group (P> 0.05). Conclusion PF4 can protect mice bone marrow hematopoietic stem cells from damage caused by ionizing radiation.
