目的研究醋酸铅致淋巴细胞的DNA损伤情况,了解铅的遗传毒性。方法用不同浓度的醋酸铅分别对大鼠进行腹腔注射体内染毒24 h及取正常大鼠血淋巴细胞进行体外染毒1 h,然后,采用单细胞凝胶电泳技术分别测定各血淋巴细胞DNA损伤程度。结果体内实验组见淋巴细胞边缘不光滑,毛糙,体外实验各处理组都有明显彗尾形成,体内外实验各染毒组彗星出现率、DNA迁移长度均明显高于阴性对照组。结论醋酸铅对大鼠淋巴细胞DNA具有一定的损伤作用。
Objective To study the DNA damage caused by lead acetate in lymphocytes and to understand the genotoxicity of lead. Methods The rats were injected intraperitoneally with different concentrations of lead acetate for 24 h and the normal rat blood lymphocytes were exposed to the virus for 1 h. The single cell gel electrophoresis was used to determine the DNA damage of each blood lymphocyte degree. Results The in vivo experimental group showed that the margins of lymphocytes were not smooth and rough, and all the treatment groups had obvious comet tail formation in vitro. The incidence of comet and the length of DNA migration in each group were significantly higher than those in the negative control group. Conclusion Lead acetate has certain damage on rat lymphocyte DNA.
